If you're using split config edit /etc/exim4/main/{something}
(personally I'd make a new file rather than editing on of the supplied
ones, that way upgrades are smoother)
If you're not using split config edit /etc/exim4/exim4.conf.template
Yes, you need to restart (or reload is sufficient) exim4. the your new
config will be compiled and syntax checked and if it passes installed in
/var/lib/exim4/config.autogenerated if not you'll get an error message.
