NginX is a great proxy, but I've never used it for SMTP (or even IMAP,
which is its oldest mail feature).

There is always GNU Anubis, which is generally expecting to be an SMTP
proxy for MUA submission, but should be easily extended (as it has a set
of capable scripting hooks) to do what you would like.
