If you're not worried about rejecting the message at SMTP time, you could bump the global limit up to take care of your two users and rely on a local mail delivery agent to enforce size limits to the rest of the user base. This idea doesn't sound very economical considering its only 2 users out of a 1000 user base. Maybe a compromise somewhere in the middle? Generally speaking mail over the 50-75 MB size tends to choke up many mail systems, and I agree with the other responder that maybe transferring files using some other method would be better.
