Might be that interpolated files are not allowed for the domain
pattern in a route_list, because this is not strictly a domain list.
It accepts only the same syntax as a *single* item in a domain list.
Whether a single item includes interpolated files is beyond my
knowlegde.

Try to use a lookup for the domains. That way, you will also be able
to specify wildcarded domains like *.t-online.de (if needed):

>         route_list = partial-lsearch;/etc/exim/routed-domains mail.example.com bydns_a


Put your routed domains in the file, on domain or pattern per line.
