You still didn't say how you aare using that macro, but I assume it's
for a hostlist. The syntax for hostlist elements is given in
https://exim.org/exim-html-current/doc/html/spec_html/ch-domain_host_address_and_local_part_lists.html#SECThostlist
Note that there are no [] around an IPv6 address.
Also, be careful of the colons in one.
