No, leave that part alone. Instead create (if it doesn't already exist) a
file /etc/exim4/exim4.conf.localmacros (for single-file) or
e.g. /etc/exim4/conf.d/main/00_localmacros (for split-file config) and add
the macro setting there:

CHECK_DATA_LOCAL_ACL_FILE = /etc/exim4/local_acl
