Just use rgrep to find out:

zsh % rgrep dnslookup /etc/exim4/conf.d/

/etc/exim4/conf.d/router/200_exim4-config_primary:dnslookup_relay_to_domains:
...

After you have changed you configuration, remember to run
update-exim4.conf.
