You're fine having them direct to root, however you will want to make
sure that root then points to a valid user, and if the rest of the
aliases are user configurable you may in fact want to point it at a full
email address so they can't just redirect the mail for your user.
As for the aliases file, Exim does not require newaliases to be run, it
reads the aliases files directly each time it needs to do the lookup (at
least for the router you pasted). Which means that unlike Sendmail you
can changes the aliases file without needing to restart Exim.
