But it would no longer be doing what it's name implies:

match the *domain* (only)

Adding prefixes to make a 'subdomain' changes the 'domain' not at all,
and being able to find *only* that, with or without prefixes, is important.

Exim has several ways of handling the longer string, or even just
selected parts of it.
