No, that doesn't reverse the polarity, unless it's followed by a reject all
acl. The test below is fairly cheap, so if you want to do more expensive
tests like sender verification callouts, they should follow that test.

You should do sender verification callouts after getting the recipient, so
that you can accept mail to postmaster.
