Sure. Several possible ways of dealing with it.
(1) Directly in an ACL for RCPT:
deny recipients = /the/file
(2) Use verify in the ACL
deny !verify = recipient
and ensure the verify fails by setting up this router
fail_router:
driver = redirect
allow_fail
data = ${lookup{$local_part@$domain}lsearch{/the/file}\
{:fail:Unknown address}}
No doubt there are other ways as well.
