You can use a seperate router like this (untested):
routingtable:
driver = manualroute
transport = remote_smtp
route_data = ${lookup{$domain}dbm{/etc/exim/routingtable}}
host_find_failed = defer
no_more
/etc/exim/routingtable has the format:
domain host_list
Example:
domain.tld: mail.domain.tld
*.domain.tld: mail.domain.tld
