You need to read the manual, your syntax is wrong.

It sees it as:
command arg0 /opt/mail2news/mail2news
        arg1 ${lookup
        arg2 {$header_list-post}lsearch{/etc/exim/mailinglists}}


Should be:
command = /opt/mail2news/mail2news "${lookup {$header_list-post:}lsearch{/etc/exim/mailinglists}}"
