This means that if you create /path/to/vdoms/files/virtual_domains and
stick in domain mydomain.net, you cna then create
/path/to/vdoms/files/mydomain.net.aliases which contain aliases for
that domain.

This sounds like a manual route:
masqueraded_hosts:
driver = manualroute
domains = ! +local_domains
route_data = ${lookup{$domain}partial-lsearch{/path/to/masqueraded_hosts}}
transport = remote_smtp
the file /path/to/masqueraded_hosts contains:
*.foo.com mail.foo.com:mail2.foo.com byname
*.foobar.org mx01.foobar.org:mx02.foobar.org byname
