You can place a catch_all router after the router which is handling
your virtual users:

catch_all:
    driver = redirect
    data = postmaster@${domain}

Yes, it is possible (mysql-support must be compiled in). An example
could be:

virtual_user:
    driver = redirect
    allow_fail
    allow_defer
    data = ${lookup mysql{ SELECT maildir FROM users \
        WHERE id='${local_part}@${domain}' }}
    directory_transport = address_directory
