Yes. Just make sure you set local_interfaces global option as well as
interface option with smtp transport.

Like:

In exim.conf:

local_interfaces = 1.2.3.4

[...]

remote_smtp:
    driver = smtp
    interface = 1.2.3.4

In exim2.conf:

local_interfaces = 1.2.3.5

[...]

remote_smtp:
    driver = smtp
    interface = 1.2.3.5
