No. INCLUDE is for included header files, not for libraries. The kind of
setting required is usually something like INCLUDE=-I/usr/local/include
or a variation.

I built Exim 3.22 on Redhat 6.2 with this Local/Makefile:

BIN_DIRECTORY=/home/ph10/exim/bin

COMPRESS_COMMAND=/opt/gnu/bin/gzip
COMPRESS_SUFFIX=gz
ZCAT_COMMAND=/opt/gnu/bin/zcat

CONFIGURE_FILE=/home/ph10/exim/configure

DBMLIB=/usr/lib/libdb.a

DIRECTOR_ALIASFILE=yes
DIRECTOR_FORWARDFILE=yes
DIRECTOR_LOCALUSER=yes
DIRECTOR_SMARTUSER=yes

EXICYCLOG_MAX=10

EXIM_GID=1169
EXIM_UID=1169

EXIM_MONITOR=eximon.bin

EXTRALIBS=-lpam -ldl

HAVE_IPV6=yes

LOOKUP_DBM=yes
LOOKUP_LSEARCH=yes

ROUTER_DOMAINLIST=yes
ROUTER_IPLITERAL=yes
ROUTER_LOOKUPHOST=yes
ROUTER_QUERYPROGRAM=yes

SPOOL_DIRECTORY=/home/ph10/exim/spool
SUPPORT_PAM=yes

TRANSPORT_APPENDFILE=yes
TRANSPORT_AUTOREPLY=yes
TRANSPORT_PIPE=yes
TRANSPORT_SMTP=yes

# End of EDITME
