Probably.

No. This is usually given as a permanent error, i.e. a 5xx return code
(usually 550), so Exim won't try again anyway. The retry rules are not
involved.

But of course, if it's a bounce message, it can't return it to sender,
so it remains on the queue. You need to set ignore_errmsg_after if you
want it to be thrown away.
