If you want the whole rule to be done by lookup, you need something like
*@* ${lookup{$domain}lsearch{/usr/whatever}{$local_part@$value}fail} Tbc
where the file contains lines like
atlantic-platinum.com atlanticplatinum.demon.co.uk
