I do this by creating a router as below:

split_route:
        driver =  manualroute
        no_check_local_user
        route_data = 
${lookup{$local_part@$domain}lsearch{/etc/exim/splitroutes}}
        ignore_target_hosts = 0.0.0.0 : 127.0.0.0/8
        transport = remote_smtp



then on the file /etc/exim/splitroutes, i have

user1@???:    IP-of-mail1.domain.com
user2@???:    IP-of-mail1.domain.com
user3@???:    IP-of-mail2.domain.com
user4@???:    IP-of-mail2.domain.com
user5@???:     IP-of-mail3.domain.com
user6@???:    IP-of-mail3.domain.com



On mail1.domain.com, I omit the entries for localhost, and same with the
others respectively
